Loss of proteostasis

The cellular housekeeping that slows down with age.

Proteostasis is the set of mechanisms that keep cellular proteins in good shape: folding them correctly, repairing those that deform, clearing those beyond saving. With age this maintenance machinery loses efficiency.

Misfolded proteins then build up and aggregate. This sits at the heart of neurodegenerative diseases such as Alzheimer's and Parkinson's, but it affects every tissue.

The scope

What this pillar covers

Autophagy

The cell's internal recycling system: how it works, what triggers it, and why it dulls with age.

Fasting and autophagy

Intermittent fasting, fasting-mimicking diets, spermidine: the best-documented triggers of cellular clean-up.

Heat shock proteins

Sauna, heat exposure, intense exercise: how HSPs protect protein structure.

Aggregates and the brain

Beta-amyloid, tau, alpha-synuclein: the link between failed clearance and cognitive decline, and the role of the nighttime glymphatic system.

What is at stake

Why it matters

A cell that stops cleaning gets cluttered. Faulty proteins accumulate, disrupt internal function, then form toxic aggregates. It is a silent mechanism that advances for decades before the first symptoms.

It is also one of the hallmarks where simple actions do the most: spacing meals, heat exposure, deep sleep. Three accessible levers that switch the clean-up machinery on directly.

A healthy cell is not one that produces a lot. It is one that knows how to throw out what is no longer useful.
Taking action

Where to start

1

Create fasting windows

A 12 to 16 hour overnight gap, held consistently, beats long and rare fasts.

2

Use heat

Sauna, hot bath, sustained exercise: thermal hormesis activates heat shock proteins.

3

Secure deep sleep

The brain clears its protein waste at night. No supplement replaces that phase.
